Description
SS-31, also written SS31, has only four residues, and two of them are unusual. Position 1 is D-arginine, the mirror-image (D) form of arginine, and position 2 is Dmt, a tyrosine carrying two extra methyl groups on its aromatic ring. Lysine and phenylalanine follow, and the C-terminus is amidated (-NH2) rather than left as a free acid.
The residues alternate between basic and aromatic: D-Arg and Lys at positions 1 and 3, Dmt and Phe at positions 2 and 4. That pattern is what "aromatic-cationic" means. Counting the free N-terminal amine, SS-31 has a net charge near +3 at pH 7, with no carboxylate on the amidated end to offset it. In a mass spectrum its [M+H]+ ion appears at m/z 640.39 (monoisotopic mass 639.39 Da).
